芍药甘草汤含药血清对纤维环细胞MyD88、IκB、NF-κB蛋白表达的影响

摘要
目的:观察芍药甘草汤含药血清对颈椎间盘纤维环细胞MyD88、IκB、NF-κB蛋白表达的影响。方法:体外培养颈椎间盘纤维环细胞,共4组,分为空白组、模型组(LPS 10ng/mL)、抑制剂组(Triptolide 10μmol/mL干预1h后加入LPS 10ng/mL)、芍药甘草汤组(芍药甘草汤含药血清0.308g/mL+LPS10ng/mL),干预24h。采用Western blot检测颈椎间盘纤维环中MyD88、IκB、NF-κB蛋白表达变化情况。结果:颈椎间盘纤维环细胞中MyD88、NF-κB P65的蛋白表达:模型组明显高于空白组(P<0.05),芍药甘草汤组明显低于模型组(P<0.05);颈椎间盘纤维环细胞IκB的蛋白表达:模型组明显低于空白组(P<0.05),芍药甘草汤组明显高于模型组(P<0.05)。结论:芍药甘草汤可以降低颈椎间盘纤维环细胞MyD88、NF-κB P65的蛋白表达量,同时上调IκB的蛋白表达量。
        Objective:To observe the Effect of Shaoyaogancao Decoction serum on the expression of MyD88,IκB and NF-κB in the annulus fibrosus cells.Methods:Cultured annulus fibrosus cells were divided into blank group,model group(LPS 10 ng/mL),inhibitor group(Triptolide 10μmol/mL after intervention 1 h was added to LPS 10 ng/mL),Shaoyaogancao Decoction group(Shaoyaogancao Decoction serum containing medium dose group+LPS 10 ng/mL),intervened for 24 hours.Results:The protein expression of MyD88 and NF-κB P65 in the annulus fibrosus cells was significantly higher in the model group than in the blank group(P<0.05).The Shaoyaogancao Decoction group was lower than the model group(P<0.05);the expression of the IκB protein in the annulus fibrosus cells:a model The group was significantly lower than the blank group(P<0.05),and the Shaoyaogancao Decoction group was significantly higher than the model group(P<0.05).Conclusion:The results showed that Shaoyaogancao Decoction can reduce the protein expression of MyD88 and NF-κB P65 and up-regulate the expression of IκB protein.
