摘要
目的探讨狼疮肾炎患者血清miRNA-374a的表达情况及其意义。方法运用Target Scan靶基因预测分析软件等生物信息学方法,预测miRNA-374a靶基因并进行分析;采用qRT-PCR方法检测miRNA-374a、MCP-1在狼疮肾炎患者血清和健康对照组血清的表达变化,并分析其与临床参数的关系。结果生物信息学分析显示MCP-1为狼疮肾炎的靶基因;与健康对照组相比,miRNA-374a在狼疮肾炎患者血清低表达(P<0.05),MCP-1在狼疮肾炎患者血清高表达(P<0.05);miRNA-374a、MCP-1的表达水平与年龄没有明显关系(P>0.05),与临床分期有关(P<0.05)。结论 miRNA-374a在狼疮肾炎患者血清中低表达,可能通过对其下游靶基因MCP-1的调控,在狼疮肾炎发生过程中发挥作用。
        Objective To investigate serum mi RNA-374 a expression in patients with lupus nephritis and its clinical significance. Methods Target genes of mi RNA-374 a were predicted by bioinformatics methods such as Target Scan and closely related gene with lupus nephritis were screened. The expression levels of serum mi RNA-374 a and MCP-1 in patients with lupus nephritis and healthy controls were detected using q RT-PCR,and the correlations between those and clinical parameters were analyzed. Results Bioinformatics analysis revealed that of mi RNA-374 a target genes was MCP-1 in lupus nephritis. Compared with healthy controls,serum mi RNA-374 a was significantly lowly expressed in patients with lupus nephritis while MCP-1 was significantly highly expressed(P<0.05);expression levels of mi RNA-374 a and MCP-1 showed no significant correlation with age(P>0.05),but with clinical stages(P<0.05). Conclusion Low expression of mi RNA-374 a was detected in the serum of patients with lupus nephritis. It may play an important role in the pathogenesis and progression of lupus nephritis by regulating the downstream target gene MCP-1.
