摘要
目的:研究穴位注射疗法对脑瘫幼鼠脑组织神经细胞及肢体运动功能和学习记忆能力的影响并探讨相关机制。方法:采用颈动脉夹断法制备30只7日龄SD脑瘫幼鼠模型并随机分为3组:模型对照组、针灸组、穴位注射组,每组10只,另设10只7日龄SD健康幼鼠为正常对照组。穴位注射组分别于幼鼠左侧顶颞前斜线和右肢穴位处注射药物,针灸组在相同部位进行针刺。治疗结束后通过悬吊实验、斜坡实验、拒俘反应实验、Morris水迷宫实验等进行神经行为学的疗效评价。取各组幼鼠脑组织进行HE染色,计录正常神经细胞的数量,并检测海马区脑组织及血清中神经元特异性烯醇化酶(neuron-specific enolase,NSE)的含量。结果:穴位注射组各神经行为学指标均优于模型对照组及针灸组,差异有统计学意义(P <0. 05);与模型对照组和针灸组相比,穴位注射组脑组织中正常神经细胞数量较多,海马区NSE含量较高而血清中NSE水平降低。结论:穴位注射疗法可以有效提高脑瘫幼鼠的肢体运动和学习记忆能力,且效果优于单纯针灸疗法,其机制可能是在穴位与药物双重作用下,受损神经元得以修复,减少了NSE向血液中的渗漏,从而有效地改善脑瘫状态。
        Objective: To study the effect of acupoint injection therapy on the motor function and learning and memory ability of cerebral nerve cells and limbs in infant rats with cerebral palsy and to explore the possible mechanism. Methods: 30 7-day-old SD cerebral palsy rats models were prepared by carotid artery clamping method and randomly divided into three groups: model control group,acupuncture group and acupoint injection group,10 in each group,and 10 healthy SD rats aged 7 days were set as normal control group. Acupoint injection group injected drugs at left parietal-temporal anterior oblique line and right limb acupoints respectively. Acupuncture group received acupuncture at the same location. After the treatment,the neurobehavioral effects were evaluated by suspension test,slope test,captive rejection test,morris water maze test and so on. HE staining was used to count the number of normal nerve cells in the brain tissues of young rats in each group,and the content of neuron-specific enolase( NSE) in hippocampus brain tissues and serum was detected. Results: The neurobehavioral indexes of acupoint injection group were better than those of model control group and acupuncture group. The difference was statistically significant( P < 0. 05). Compared with model control group and acupuncture group,the acupoint injection group had more normal neurons in brain tissue,higher NSE content in hippocampus and lower NSE level in serum. Conclusion: Acupoint injection therapy can effectively improve limb movement and learning and memory ability of cerebral palsy young rats,and its effect is better than that of acupuncture alone. The mechanism may be that under the dual action of acupoints and drugs,damaged neurons can be repaired,the leakage of NSE into blood is reduced,and the state of cerebral palsy can be effectively improved.
