摘要
目的建立T细胞受体脱氧核糖核酸重排删除环(TREC)的检测方法,并分析TREC与艾滋病疾病进展之间的关系。方法利用实时定量聚合酶链反应(PCR)技术,建立检测TREC水平的方法,并对未治疗病人及治疗后免疫重建成功与失败病人外周血中TREC水平进行分析。结果 85例未治疗的艾滋病病毒(HIV)感染者,CD4~+T淋巴细胞(简称CD4细胞)>350个/μL病人的TREC水平(32.5±7.65)高于CD4细胞在200~350个/μL的病人(19.9±6.26),而CD4细胞<200个/μL病人的TREC水平最低(9.5±1.87);免疫重建成功病人的TREC水平(28.2±10.1)远高于免疫重建失败病人(10±5.1)。结论未治疗的HIV感染者,随着CD4细胞水平的增加,外周血中TREC细胞的水平也明显增加;免疫重建成功病人外周血中TREC的水平明显高于免疫重建失败的病人。
        Objective To establish a method for detection of TREC and to analyze the relationship between the levels of TREC in peripheral blood and the progression of HIV infection. Methods We established real-time PCR method to detect TREC levels in peripheral blood of untreated patients and patients with successful or failed immune reconstitution after treatment and compared the differences of TREC between different patients. Results In untreated patients with the increased CD4, TREC level was significantly increasing. The level of TREC was significantly higher in the successful immune reconstitution patients than that in the patients of immune reconstitution failure. Conclusion The level of TREC cells in peripheral blood increases significantly with the increase of CD4 cell level in untreated patients. The level of TREC in peripheral blood of successful immune reconstruction patients is significantly higher than that of patients with failed immune reconstruction.
