衡阳地区怀化秤锤树苗木繁殖技术

摘要
为加快怀化秤锤树在衡阳的栽培与利用,引种怀化秤锤树进行了种子繁殖、扦插繁殖、组培繁殖等技术研究,并总结了3种繁殖技术。怀化秤锤树种子具有深休眠特性,须经沙藏越冬后,翌年春季3—4月播种。树枝条扦插的最佳时段为5月中下旬至6月下旬,选择3~5 a树龄的当年生半木质化健壮枝条作为插穗,采用黄泥、河沙、泥炭(体积比为2∶1∶1)配制混合基质,扦插后2周覆膜保温保湿,成活率85%以上。组培繁殖时采集当年生未木质化嫩枝的单芽茎段接种在培养基MS+BA 1.0 mg/L+NAA 0.1 mg/L+蔗糖30.0 g/L+琼脂6.0 g/L上,培养25~30 d后,可长出3~5个芽,接着转移到培养基MS+BA 2.0 mg/L+NAA 0.2 mg/L+蔗糖30.0 g/L+琼脂6.0 g/L上进行继代增殖培养,30 d后增殖倍数可达3~4倍,将继代增殖芽苗转接到培养基1/2 MS+IBA 0.5 mg/L+NAA 0.2 mg/L+蔗糖30.0 g/L+琼脂6.0 g/L上进行生根培养,20 d后产生不定根,生根率达95%以上。
        Huaihua Sinojackia xylocarpa has been listed in the second batch of the List of Key Wild Plants under State Protection. In order to speed up the cultivation and utilization of Huaihua Sinojackia xylocarpa in Hengyang, the introduction of Huaihua Sinojackia xylocarpa was studied on seed propagation, cuttage propagation and tissue culture propagation. The seeds of Huaihua Sinojackia xylocarpa had the characteristics of deep dormancy. It had to be sown in the following spring from March to April after winter in sand storage. The optimum time for cutting branches was from mid-late May to late June. Half-lignified strong branches of 3-5 years old were selected as cuttings.Mixed substrates of yellow peat, river sand and peat(volume ratio was 2 ∶ 1 ∶ 1) were used to prepare mixed substrates. After cutting, film was covered for two weeks for heat preservation and moisture retention, survival rate was more than 85%. The single bud segments of the non-lignified tender shoots were collected and inoculated on the culture medium(MS+BA 1.0 mg/L+NAA 0.1 mg/L+ sucrose 30.0 g/L+agar 6.0 g/L). After 25-30 days of culture, 3-5 buds could grow. Then transferred to the culture medium(MS+BA 2.0 mg/L+NAA 0.2 mg/L+ sucrose 30.0 g/L+ agar 6.0 g/L) for subculture. After 30 days, the proliferation multiple could reach 3-4 times. Transfer of subcultured proliferating bud seedlings to medium(1/2 MS+IBA 0.5 mg/L+NAA 0.2 mg/L+sucrose 30.0 g/L+agar 6.0 g/L) for rooting culture, and adventitious roots were produced after 20 days, and the rooting rate was over 95%.
