摘要
目的建立高效液相色谱法(HPLC)分析消癌平注射液中通关藤苷A和I的方法并测定其含量。方法消癌平注射液经微孔滤膜过滤,用HPLC进行分析,采用Symmetry C18(5μm,250 mm×4. 6 mm)色谱柱;流动相:乙腈(A)-0. 1%磷酸水溶液(B);梯度洗脱方式(0~10 min,32%A; 10~10. 01 min,32%A~42%A; 10. 01~15 min,42%A; 15~15. 01 min,42%A~50%A; 15. 01~28 min,50%A);流速:1. 0 ml/min;检测波长:223 nm。结果通关藤苷A和I的线性范围分别为6. 60~132. 00、3. 52~70. 40μg/ml,通关藤苷A和I的平均加样回收率均> 98%,相对标准偏差均<3%。结论 HPLC准确可靠、操作简便,可用于同时测定消癌平注射液中通关藤苷A和I的含量。
        Objective To establish an high performance liquid chromatography( HPLC) method for detection of Caulis Marsdeniae Tenacissimae glycoside A and I contents in Xiaoaiping Injection. Methods Xiaoaiping Injection was filtrated by microporous membrane and analyzed by HPLC. Chromatographic separation was performed on Symmetry C18( 5 μm,250 mm × 4. 6 mm) column,and the mobile phase was composed of acetonitrile( A) and-0. 1% phosphoric acid aqueous solution at a 1. 0 m L/min flow rate,and gradient elution methods were 32% A within 0-10 min,32% A-42% A within 10-10. 01 min,42% A within 10. 01-15 min,42% A-50% A within 15-15. 01 min and 50% A within 15. 01-28 min,and the detection wavelength was 223 nm. Results Calibration curves of Caulis Marsdeniae Tenacissimae glycoside A and I were linear in ranges of 6. 60-132. 00 and 3. 52-70. 40 μg/ml respectively. Average recoveries of Caulis Marsdeniae Tenacissimae glycoside A and I were all larger than 98%,and values of relative standard deviation( RSD)were all less than 3%. Conclusion HPLC method is reliable,simple and accurate which can be used for simultaneous detection of Caulis Marsdeniae Tenacissimae glycoside A and I contents in Xiaoaiping Injection.
