静宁颗粒对注意力缺陷多动障碍模型大鼠血清单胺类神经递质的影响

摘要
目的探讨静宁颗粒治疗注意力缺陷多动障碍(ADHD)可能的作用靶点。方法选用雄性自发性高血压(SHR)大鼠50只,随机分为空白模型组,盐酸托莫西汀组,静宁颗粒低、中、高剂量组,每组10只。同时,将10只同龄Wistar大鼠设为正常对照组。静宁低、中、高剂量组分别给予5. 785 g/kg、11. 57 g/kg、23. 14 g/kg静宁颗粒灌胃,盐酸托莫西汀组给予盐酸托莫西汀5. 36 mg/kg灌胃,空白模型组及正常对照组给予1 ml/100 g生理盐水灌胃。各组于治疗6周后处死大鼠,采用腹主动脉取血的方法取各组大鼠血清,并采用高效液相-电化学法测定其多巴胺(DA)、去甲肾上腺素(NE)及5-羟色胺(5-HT)的含量。结果空白模型组血清中DA、5-HT均低于正常对照组(P <0. 01);静宁颗粒高、中剂量组血清中DA、5-HT含量高于空白模型组(P <0. 05,P <0. 01);静宁中剂量组中血清DA、5-HT含量高于盐酸托莫西汀组及静宁颗粒低剂量组(P <0. 05)。结论静宁颗粒可以提高血清中DA、5-HT的含量,可能是治疗ADHD的作用靶点之一,以中剂量为宜。
        Objective To explore the potential effects of jingning granules on attention deficit hyperactivity disorder( ADHD). Methods A total of 50 male rats with spontaneous hypertension were randomized into a blank model group,an atomoxetine hydrochloride group,the low-dose group,the middle-dose group and the high-dose group of jingning granules,10 rats in each one. Simultaneously,10 Wistar rats of the same age were selected as the normal control group. The gavage with jingning granules was used in the low-dose group,the middle-dose group and the high-dose group of jingning granules,with 5. 785 g/kg,11. 57 g/kg and 23. 14 g/kg successively. In the atomoxetine hydrochloride group,the gavage was given with atomoxetine hydrochloride,5. 36 mg/kg. In the blank model group and the normal control group,the gavage was given with physical saline,1 ml/100 g. After treatment for 6 weeks,the rats were sacrificed in each group. The serum sample was collected from abdominal aorta. The high performance liquid-electrochemistry method was adopted to determine the levels of dopamine( DA),norepinephrine( NE) and 5-hydroxytryptamine( 5-HT). Results The levels of serum DA and 5-HT in the blank model group were lower than the normal control group( P < 0. 01). In the high-dose group and the middle-dose group of jingning granules,the levels of serum DA and 5-HT were higher than the blank model group( P < 0. 05,P < 0. 01). In the middle-dose group of jingning group,the levels of serum DA and 5-HT were higher than the atomoxetine hydrochloride group and the low-dose group of jingning granule( P < 0. 05). Conclusion Jingning granules improve the levels of serum DA and 5-HT,which is probably one of the effect mechanisms in the treatment of ADHD. The best effect is achieved with the middle-dose jingning granules.
