光照强度对Kisspeptin及其受体GPR54在鸡脑表达的影响

摘要
随机选用100日龄京红Ⅰ号蛋鸡,分别接受光照强度30,20,10,1 lx处理,运用免疫组化技术测定Kisspeptin和GPR54免疫反应阳性神经元在鸡中脑和间脑的分布和表达。结果显示:Kisspeptin免疫反应阳性神经元主要分布于中脑SGC、Imc、Ipc、顶盖前核(Pt)、外侧螺旋核(SPL)。在SGC、Imc、Ipc,平均灰度值和面积百分比,30,20 lx组显著高于10,1 lx组(P<0.05),在Pt和SPL,30 lx平均灰度值和面积百分比最高。GPR54免疫反应阳性神经元主要分布于中脑的lmc、Ipc、SP/IPS、nbor、SGC,平均灰度值和面积百分比,光照强度30,20 lx组显著大于l0,1 lx组(P<0.05)。Kisspeptin免疫反应阳性神经元主要分布于间脑的下丘脑后内侧核(MPH)、丘脑室旁核前部(PVa)、PHN、PVN、Rot。在Rot、PHN、PVN和PVa,Kisspeptin免疫反应阳性神经元平均灰度值和面积百分比,30,20 lx组显著大于l0,1 lx组(P>0.05);在MPH,30 lx高于20 lx,20 lx高于10 lx,10 lx高于1 lx组。GPR54免疫阳性神经元主要分布于鸡间脑PVN、GLv、Rot、PHN和CPa。平均灰度值和面积百分比在Rot,30 lx组最大,但各组间差异不显著(P>0.05),在PVN、PHN、GLv和CPa,30,20 lx组显著大于10,1 lx组(P<0.05)。Kisspeptin免疫反应阳性细胞在腺垂体的平均灰度值,光照强度20 lx组最大,面积百分比30 lx最大,但差异不显著(P>0.05)。结果表明,光照强度能促进Kisspeptin和GRP54在蛋鸡中脑和间脑表达,随光照强度增加,表达量增加。Kisspeptin接受光信息调控,通过调节GnRH分泌,参与调节鸡的生殖机能。
        Jinghong-I hens aged 100 d were randomly divided into four groups and applied a light of 30,20,10,1 lx respectively,to mensurate the distribution and expression of Kisspeptin and GPR54 immunoreactive positive neurons in hen mesencephalon and diencephalon using immunohistochemical method.The results showed that Kisspeptin immunoreactive positive neurons were mainly observed in SGC,Imc,Ipc,Pt,and SPL of mesencephalon.The mean gray values and percent area of positive neurons in 30 lx and 20 lx groups were significantly higher than those of 10 lx and 1 lx groups(P<0.05) in SGC,Imc,and Ipc.That in Pt and SPL was the largest of 30 lx group among the four groups.GPR54 immunoreactive positive neurons were mainly seen in Imc,Ipc,SP/IPS,nbor,and SGC of mesencephalon.The mean gray values and percent cell area of positive neurons in 30 lx and 20 lx groups were significantly higher than those of 10 lx and 1 lx groups(P<0.05).Kisspeptin immunoreactive positive neurons were mainly observed in MPH,PVa,PHN,PVN,and Rot of diencephalon.The mean gray values and percent area of positive neurons in 30 lx and 20 lx were significantly higher than those of 10 lx and 1 lx groups(P<0.05)in Rot,PHN,PVN,and PVa.The mean gray values and percent area of positive neurons depend on light intensity in MPH.GPR54 immunoreactive positive neurons were mainly seen in PVN,GLv,Rot,PHN,and CPa of diencephalon.The mean gray values and percent area in Rot of 30 lx group was the highest,without significant difference(P>0.05)among the four groups.That in PVN,PHN,GLv,and Cpa of 30 lx and 20 lx group were significant higher than 10 lx and 1 lx.It suggested that with the increase of light intensity,the expression of Kisspeptin and GRP54 increase significantly in hen mesencephalon and diencephalon.Kisspeptin receives light information and regulates the reproductive function of chicken by regulating the secretion of GnRH.
