摘要
目的:测定国产血竭、进口血竭药材中10种微量元素含量,研究各微量元素的分布规律和相关性。方法:微波消解法处理样品,利用火焰原子吸收法和石墨炉原子吸收法测定样品中Fe、Cu、Zn、Mg、Ca、Sr、K、Pb、Cd、Cr共10种微量元素含量,通过Radar图分析法和Spearman等级相关系数解析2种血竭中各元素分布规律和相关性。结果:元素加标回收率在96.7%~104.9%范围,RSD<2%。Radar图分析发现,Fe、K、Ca在2种血竭中分布最为广泛(0~4 000μg·g~(-1)),Cu、Mg、Sr、Zn次之(0~140μg·g~(-1)),进口血竭中K、Ca、Fe、Mg、Cu含量高于国产血竭,而Sr、Zn含量较低。国产血竭中有较高含量的重金属元素Cr,重金属元素Pb、Cd在2种血竭中分布相当(0~0.10μg·g~(-1));Spearman相关性分析表明,国产血竭中Cu-Fe、CuZn、Fe-Zn相关系数高达0.9以上水平(P<0.01),而进口血竭中,Cr-Fe、Cr-Mg、Cr-Zn则呈现极显著负相关(P<0.01)。结论:本研究建立的元素测定方法准确、可靠,2种血竭中元素分布具有规律性,药效相关元素间呈现不同程度的相关性,可以为2种血竭的临床使用和药效分析研究提供参考。
        Objective:To determine and investigate the distribution and correlation of 10 trace elements in Draconis Resina and Draconis Sanguis. Methods:Samples were processed by microwave digestion. The content of 10 trace elements,Cu,Zn,Fe Mg,Ca,Sr,K,Pb,Cd and Cr,were determined by flame atomic absorption spectrometry and graphite furnace atomic absorption spectrometry. The distribution and correlation of elements in two kinds of "dragon's blood" were analyzed by Radar chart and Spearman rank correlation coefficient. Results:Element recoveries were in the range of 96.7%-104.9% with RSDs<2%. Radar analysis showed that Fe,K and Ca were most widely distributed in two kinds of "dragon's blood"(0-4 000 μg·g~(-1)),followed by Cu,Mg,Sr and Zn(0-140 μg·g~(-1)). The contents of K,Ca,Fe,Mg and Cu in the Draconis Sanguis were higher than those in Draconis Resina,while the contents of Sr and Zn were lower. There was a higher content of heavy metal elements Cr in Resina Draconis,and heavy metal element Cd,Pb were distributed equivalently in two"dragon's blood"(0-0.10 μg·g~(-1));Spearman correlation analysis showed that the correlation coefficients of Cu-Fe,Cu-Zn and Fe-Zn in Resina Draconis were more than 0.9(P<0.01),while Cr-Fe、Cr-Mg and Cr-Zn in Draconis Sanguis showed significant negative correlations(P<0.01). Conclusion:The method of element determination established in this study is accurate and reliable. The distribution of elements in two kinds of"dragon's blood" is regular,and the elements correlating to drug efficacy have different degrees of correlation,which provides an important basis for the clinical use and pharmacodynamic analysis of two kinds of "dragon's blood".
