逆电针少阳经特定穴对偏头痛大鼠50%缩足阈值及血清中CGRP、SP含量的影响

摘要
目的观察逆电针少阳经特定穴对偏头痛大鼠的50%缩足阈值(50%PWT)及血清中降钙素基因相关肽(CGRP)、P物质(SP)含量的影响,探讨其可能的作用机制。方法将60只大鼠随机分为空白组(n=20)、模型组(n=20)、逆电针组(n=20),每组又分为造模后30 min组、造模后90 min组,每组各10只。空白组、模型组正常饲养并同电针组同时段捆绑30 min×7 d,逆电针组正常饲养并捆绑行电针治疗30 min×7 d。空白组第8天皮下注射等量0.9%氯化钠注射液,模型组、逆电针组第8天皮下注射硝酸甘油造模,观察治疗前、造模后大鼠行为学、50%PWT,酶联免疫吸附法(ELISA)检测各组血清中CGRP、SP的含量。结果与空白组比较,造模后模型组大鼠50%PWT明显降低(P <0.01),行为学评分及CGRP、SP含量升高(P <0.01);与模型组比较,逆电针组造模后50%PWT明显升高(P <0.01),CGRP、SP含量下降(P <0.01);与造模后30 min比较,模型组50%PWT造模后90 min升高(P <0.05),CGRP、SP含量无明显差异(P> 0.05),逆电针组50%PWT造模后90 min升高(P <0.05),CGRP、SP含量下降(P <0.05)。结论逆电针少阳经特定穴可以下调血清中CGRP、SP的表达,减轻大鼠偏头痛,其机制可能是通过减少三叉神经末梢对CGRP、SP等神经递质的释放,抑制三叉神经血管系统的激活,从而缓解疼痛。
        Objective: To observe the effects of pre-electroacupuncture specific acupoint on Shaoyang meridians on 50%PWT and serum calcitonin gene related peptide(CGRP),Substance P(SP) content in migraine rats,and to explore its possible mechanism. Methods: 60 rats were randomly divided into four groups: the blank group(group C),the model group(group M) and pre-electroacupuncture group(group PEA). Each group was divided into group 30 min after modeling and group 90 min after modeling,10 rats in each. The rats from group PEA were bound and given electro-acupuncture 30 min per day for 7 days. The other two groups were only bound at the same time. All rats were subcutaneously injected with nitroglycerin on day 8,while the control group was injected with the same volume of normal saline. 50%PWT were observed before and after modeling and treatment.The levels of CGRP and SP in serum were determined by enzyme-linked immunosorbent assay(ELISA). Results:Compared with the blank group,the model group significantly increased behavior score and the levels of CGRP and SP(P < 0.01),while PWT was significantly decreased(P < 0.01). However,PEA given which markedly alleviated down-regulated the levels of CGRP and SP(P < 0.01),whereas significantly increased the 50%PWT(P <0.01). Compared with the group 30 min after modeling,the model group increased 50%PWT(P < 0.05),while the the levels of CGRP and SP had no significant difference(P > 0.05) after modeling 90 minutes;the group PEA increased 50%PWT(P < 0.05),and the the levels of CGRP and SP was decreased after modeling 90 minutes(P <0.05). Conclusion: Pre-electroacupuncture specific acupoint on Shaoyang meridians can inhibit the expression of vascular active substances,such as CGRP and SP,thereby reducing the level of migraine pain,which maybe related to reducing the release of CGRP,SP and other neurotransmitters from trigeminal nerve endings;the activation of trigeminal nerve and vascular system can be inhibited,thus alleviating pain.
