四倍体野菊与露地菊‘紫秋裳’种间杂交 F_1代3个株系倍性鉴定

摘要
采用传统根尖压片技术对母本四倍体野菊(Dendrathema indicum)与父本露地菊‘紫秋裳’(Dendrathema×gandiflorum‘ziqiushang’)种间杂交F_1代3个株系(‘S05’、‘S09’、‘S10’)进行染色体计数观察,并运用流式细胞仪测定样品的荧光强度值进而鉴定其倍性。结果表明:母本四倍体野菊、父本露地菊‘紫秋裳’、‘S05’、‘S09’、‘S10’的染色体数目分别为36条、52~54条、36条、35~38条、36~38条,该试验的染色体计数结果不同于理论上由遗传规律得到的结果;利用流式细胞仪检测荧光强度峰值分别为344.98、401.36、327.64、371.75、309.41,但检测结果受外界影响,准确性有待进一步探讨。
        The chromosome counts of three F_1 strains(‘S05', ‘S09' and ‘S10') of interspecific hybridization between female tetraploid(Dendrathemaindicum) and the father of the genus(Dendrathema×gandiflorum ‘ziqiushang') were observed by using traditional root tip tablet technology, and the fluorescence intensity value of the samples was determined by flow cytometry to identify the ploidy. The number of chromosomes of the female tetraploid(Dendrathemaindicum), the father of the genus(Dendrathema×gandiflorum ‘ziqiushang'), ‘S05', ‘S09' and ‘S10' were 36, 52-54, 36, 35-38 and 36-38, respectively, and the results of chromosome count were different from those obtained theoretically by genetic law.The peak fluorescence intensity detected by flow cytometry were 344.98, 401.36, 327.64, 371.75, and 309.41, respectively, but the test results were affected by external influences, and the accuracy needed to be further explored.
