中药枳壳“宽中除胀”作用及炮制增效机制分析

摘要
目的:明确枳壳"宽中除胀"的作用机制。方法:将SD大鼠均分为空白组,生品枳壳高(10g/kg,下同)和低(1g/kg,下同)剂量组,麸炒枳壳高、低剂量组,蜜麸枳壳高、低剂量组,功能性消化不良(FD)模型组以及多潘立酮片组,每组6只,采用蛋白质免疫印迹(Western Blot)与免疫组化(IHC)法检测组织中胃泌素(GAS)和生长抑素(SS)的蛋白表达水平与阳性细胞分布情况。结果:Western Blot和IHC结果均显示模型组大鼠下丘脑和胃窦GAS表达较空白组显著降低(P<0.01),SS表达较空白组显著增加(P<0.01);各剂量给药组下丘脑和胃窦GAS、SS表达较之模型组均有不同程度上调和下调;与模型组比较,多潘立酮组下丘脑GAS、SS表达无显著性变化,而胃窦GAS、SS表达显著上调和下调(P<0.01)。枳壳各炮制品中,均以蜜麸枳壳对于FD大鼠下丘脑和胃窦GAS、SS的调节作用最佳,且低剂量组调节作用优于高剂量组。结论:枳壳经炮制后可增强宽中除胀作用,其机制可能与药物能调节下丘脑和胃窦GAS、SS蛋白表达,恢复外周胃肠激素分泌紊乱有关;麸炒品与蜜麸炒品均能有效增强生品枳壳对FD的治疗作用,且以蜜麸炒品炮制作用为佳。
        Objective: To clarify the mechanism of Fructus Aurantii in regulating vital energy and eliminating flatulence.Methods: SD rats were divided into blank group, high(10 g/kg, the same below) and low(1 g/kg, the same below) dose groups of raw Fructus aurantii, high and low dose groups of Fructus aurantii stir-frying with bran, high and low dose groups of Fructus aurantii with honey bran, FD model group and domperidone tablet group. The protein expression and distribution of positive cells of GAS and SS were detected by Western Blot and IHC. Results: Both Western Blot and IHC results showed that the expression of GAS in hypothalamus and sinuses ventriculi of model group was significantly lower than that of blank group(P<0.01), and SS expression was significantly higher than that of blank group(P<0.01). Compared with the model group, the expression of GAS and SS in hypothalamus and sinuses ventriculi of each dose group increased and decreased to some extent. Compared with the model group, the expressions of GAS and SS in hypothalamus of domperidone group had no significant difference, while the expressions of GAS and SS in sinuses ventriculi were significantly up-regulated and down-regulated(P<0.01). Among the processed products of Fructus Aurantii, Fructus Aurantii stir-frying with honey bran had the best regulating effect on GAS and SS in hypothalamus and sinuses ventriculi of FD rats, and the regulating effect of low dose group was better than that of high dose group. Conclusion:Fructus Aurantii can enhance the effect of regulating vital energy and eliminating flatulence, which may be related to the mechanism that drugs can regulate the expression of GAS and SS in hypothalamus and sinuses ventriculi, and recover the disorder of gastrointestinal hormone secretion. Products stir-frying with bran and stir-frying with honey bran both can effectively enhance the therapeutic effect of raw Fructus Aurantii on FD, and the processing effect of honey bran Fructus Aurantii is the best.
