摘要
建立小鼠体内奋乃静血药浓度液相分析方法,研究聚乙二醇修饰奋乃静缓释微球小鼠体内药动力学。采用高效液相色谱法,色谱柱为Diamonsil C18柱(250mm×4.6mm,5μm);流动相pH为3.4的缓冲液(500mL双蒸馏水中加入2mL三乙胺,用冰醋酸调节pH为3.4)-甲醇(32:68);流速0.8mL/min;柱温30℃;检测波长258nm;进样量20μL;以邻苯二甲酸二乙酯为内标物,测定小鼠体内血液中奋乃静的浓度。该方法分离度和专属性好,灵敏度高,重复性好,操作快速便捷。用聚乙二醇修饰奋乃静制备微球制剂,进行了小鼠体内药动力学研究,结果表明,聚乙二醇修饰奋乃静缓释微球制剂在小鼠体内缓释效果良好。
        In this paper,a liquid phase analysis method was established to determine blood concentration of perphenazine in mice,and the pharmacokinetics of sustained release microspheres of PEG modified perphenazine in mice was studied.During this research,high performance liquid chromatography was used,and the chromatographic column was Diamonsil C18(250 mm x 4.6 mm,5μm);mobile phase was buffer solution with pH as 3.4(Add 2 mL triethylamine to 500 mL ultrapure water and adjust pH as 3.4 with glacial acetic acid)-methanol(32:68);flow rate was 0.8 mL/min,column temperature was 30 degrees,detection wavelength was 258 nm,and the sampling volume was 20μL.Diethyl phthalate was used as internal standard to determine concentration of perphenazine in blood of mice.The method had good separation and specificity,high sensitivity,good repeatability and fast and convenient operation.The results showed the sustained release microspheres of perphenazine modified by polyethylene glycol possessed good sustained release effect in mice.
